Enemies are NPCs that the player must fight in the dungeon. Most of them deal damage, whereas some few heal other enemies or create new ones. Enemies spawn in enemy or boss rooms (except for Turret, Chester, and Mad Scientist). To pass the room, the player must defeat all enemies in it.
Enemies found in the last room of level x-5 are bosses. Bosses are more powerful enemies that block the way into the next level; a portal leading to the next level will spawn after defeating them.
Champions
Sometimes more powerful enemies who are stronger than their original form appear. These are known as Champions. They are easily identified as they are bigger and have a purple glow around them.
Enemy Generators
In some biomes, there is a type of special enemy whose function is to generate a specified list of enemies. They have these common traits:
- Enemy generators do not move.
- Before Update 3.0.0, they did not appear in additional waves.
- If there is less than a specific amount of the target enemy, the generator will spawn them until reaching the amount.
- Spawned enemies do not count in kills or drop anything, unless they are champions spawned by champion generators.
- An enemy generator seems to have a high weight of enemy capacity, to the extent that it is possible to have one room containing one generator and nothing else. At the same time, a room with common enemies will usually contain a few of them.

Trivia
- In Update 1.7.9, the rounded sniper bullets went from having a pomegranate red to a tangerine, probably for players to tell them apart.
- Before Update 1.8.2, only some of the enemies could be champions.
- From 1.8.2 to 1.8.3, enemy's oval bullets were also much smaller than normal.
- In some old versions, lines of quakes could be interrupted or reflected with melee attacks. However now they can only be reflected by Taoist's second skill.
- The enemies used to have 'rarities'/colored names, similar to weapons, in the cellar. It was removed since Update 2.7.0.
- Their names used to be White, Blue, Green, Orange and (in older versions) Red, with 'rarity' similar to those of weapons.
- All of the bosses used to have their name in Red and then it changed Orange in the Cellar before this change.
